Each delivery/pickup date has an Edit Note button (in the right column of the date section; existing notes are shown in a pale-yellow box). Click it, type into Delivery Notes (up to 100 characters) and click Save. The note is stored on that date only and shows up on the Delivery and Pickup reports and on the Uber Routes screen (for Real Time/Uber Direct deliveries).
