No. Two separate things happen: (1) When you edit an Employee Role at Employee Roles (linked from the Employees page), the POS-specific permission checkboxes always appear so you can turn them on for that role — this list isn't shown on the regular admin Roles/Profiles screen. (2) When an employee actually signs in at the POS terminal, each POS permission is only granted if both the role has it checked and the account has the POS module enabled — having the module on its own doesn't check any boxes for you. The one exception is an account flagged as a superadmin employee, which is automatically granted every POS permission regardless of what's checked.
